Research Description
Research in our laboratory is concentrated in two major areas:
1) The development and application of advanced electron and light
optical methods for the study of subcellular composition and intra- and
intercellular signal transduction and
2) the molecular mechanisms of contractile regulation and of the
cross-bridge cycle in smooth muscle.

| The map (128 x 128 at 5 seconds per pixel = 23 hours) includes part of the nucleus and a portion of the nuclear pole containing several atrial granules and mitochondria (elongated green structures). Three elements, P(green), S (yellow), and CI (blue) are shown individually. In the overlay image of Ca and P areas containing only Ca are red and only P are green, whereas areas with both elements are yellow. The intensities of the four colours were contrast enhanced to indicate the relative elemental distributions, with the weakest intensity being the lowest and the brightest intensity being the highest number of atoms (peak counts with background subtracted) of each element in the section. |
We are currently using X-ray mapping and electron probe X-ray microanalysis for determining the composition of membrane and cell organelles at 20 to 100 nm resolution, while developing energy filtered scanning transmission electron microscopy for obtaining a compositional imaging resolution of 2 to 3 nm. The latter methods are to be used for quantitatively imaging and determining the effects of calcium binding and phosphorylation on surface and organelle membranes, and on membranes mediating cell-to-cell conduction, with emphasis on the effect of calcium binding to gap junctions on cardiac conduction.
Our studies of contractile regulation are designed to identify the molecular level mechanisms of the G-protein-coupled signaling cascade, first identified by us, that inhibits myosin light chain phosphatase and so modulates the Ca2+ sensitivity of smooth muscle. A second objective is to determine how [MgADP] modulates force maintenance by unphosphorylated cross-bridges in smooth muscles containing different myosin isoforms.
The laboratory also offers a wide range of opportunities for applying high resolution compositional imaging technology (X-ray mapping, energy-filtered electron microscopy) to studies of cell signaling, subcellular ion transport and compartmentalization in cell systems ranging from bacteria through red blood cells to invertebrate photoreceptors.
